摘要:本研究通過(guò)堿提酸沉法提取黑豆蛋白,采用多酶分步協(xié)同水解黑豆蛋白,用超濾、高速逆流色譜以及凝膠過(guò)濾色譜對(duì)黑豆蛋白混合肽進(jìn)行分離純化,以膽固醇膠束溶解度抑制率為評(píng)價(jià)指標(biāo),測(cè)定黑豆蛋白混合肽及其分離純化組分的降膽固醇活性,篩選降膽固醇活性較強(qiáng)的黑豆肽組分。 黑豆肽通過(guò)超濾系統(tǒng)后,其膽固醇膠束溶解度抑制率由20.58%增大到26.57%,然后采用高速逆流色譜分離純化,收集120-140min內(nèi)洗脫下來(lái)的組分,抑制率為44.10%。降膽固醇肽組分經(jīng)凝膠過(guò)濾色譜分離得到五個(gè)峰,測(cè)定分離后各峰的膽固醇膠束溶解度抑制率,得出產(chǎn)生第三個(gè)峰的組分對(duì)膽固醇膠束溶解度的抑制活性最強(qiáng),抑制率為50.47%,其相對(duì)分子質(zhì)量為453-916之間。 關(guān)鍵字 黑豆蛋白;降膽固醇活性;分離純化
Abstract:This study by alkali and acid extraction of black soybean protein precipitation method,using multiple enzyme step hydrolysis of black soybean protein,soybean protein on mixed peptides were separated and purified by ultrafiltration,high-speed countercurrent chromatography and gel filtration chromatography,cholesterol micellar solubility inhibition rate as the evaluation index,the determination of the cholesterol-lowering activity of black bean protein component of hybrid peptide and its purification,screening cholesterol-lowering activity of black beans peptide component. Black beans peptide by ultrafiltration system,cholesterol micellar solubility inhibition rate of 20.58% increase to 26.57%,and then using the high-speed counter-current chromatography separation and purification,collection of components eluted in the 120-140min,inhibition rate was 44.10%.Cholesterol-lowering peptide component by gel filtration chromatography,five peaks of separation cholesterol micellar solubility measured after separation of each peak inhibition rate obtained generating component of the third peak is the strongest inhibitory activity on micellar solubility of cholesterol,inhibition rate was 50.47%,and the relative molecular mass of between 453-916.
