摘要:本文在實(shí)驗(yàn)室條件下,采用瓊脂平板法,研究了不同濃度的銅離子(CK、2、4、6、10、40mg/L )對(duì)菊苣(Cichorium intybus L.)種子萌發(fā)的影響。結(jié)果表明:(1)① MS培養(yǎng)基培養(yǎng)的種子,其種子發(fā)芽率和芽長(zhǎng)值隨著銅離子濃度的升高出現(xiàn)明顯的峰值;而其根長(zhǎng)值隨著銅濃度升高變化不大、峰值不明顯。②糖水培養(yǎng)基培養(yǎng)的種子,其種子發(fā)芽率和根長(zhǎng)值隨著銅離子濃度的升高出現(xiàn)明顯的峰值;而其芽長(zhǎng)值隨著銅濃度升高變化不大、峰值不明顯。 (2)①當(dāng)銅離子濃度為0mg/L-4mg/L,促進(jìn)種子發(fā)芽。當(dāng)濃度為4mg/L,種子的發(fā)芽率最高。濃度為4mg/L-10mg/L時(shí),對(duì)種子的萌發(fā)有明顯的抑制作用,萌發(fā)率迅速降低。濃度達(dá)到10mg/L及以上時(shí),種子的發(fā)芽率緩慢降低。濃度達(dá)到40mg/L時(shí)種子幾乎無(wú)法萌發(fā)②銅離子濃度為0mg/L-2mg/L時(shí),促進(jìn)種子的根和芽的生長(zhǎng)。濃度為2mg/L-10mg/L時(shí),對(duì)種子根和芽的生長(zhǎng)具有明顯的抑制作用,種子的根長(zhǎng)和芽長(zhǎng)值迅速降低。濃度達(dá)到10mg/L及以上時(shí),種子的根長(zhǎng)和芽長(zhǎng)值緩慢降低,根嚴(yán)重發(fā)育不良。 關(guān)鍵詞: 菊苣;種子;銅離子;發(fā)芽率;培養(yǎng)基
Abstract:A experiment was conducted to study the effects of different concentration(CK, 0, 2, 4, 10, and 40 mg/L) Cu2+ and different culture medium in nutrient solution on the growth of Cichorium intybus L.. The result is: (1) ①The germination rate and the number of shoot length had the maximum when the seeds grew in MS culture medium. But the number of root length changed slightly when the density of Cu2+ was different. ②The germination rate and the number of root length had the maximum when the seeds grew in water-sugar culture medium. But the number of shoot length changed slightly when the density of Cu2+ was different. (2) ①When the density of Cu2+ was 0mg/L-4mg/L , the copper ions promoted the germination rate, and there was the largest germination rate at 4mg/L of Cu2+ . When the density of Cu2+ was 4mg/L-10mg/L , the copper ions refrained germination obviously. The number of germination rate reduced quickly. When the density of Cu2+ reached 10mg/L and more, the germination rate reduced slowly. ②When the density of Cu2+ was 0mg/L-2mg/L , the copper ions promoted the number of root length and shoot length. When the density of Cu2+ was 2mg/L-10mg/L , the copper ions refrained root growth and shoot growth obviously. The number of root length and shoot length reduced quickly. The root and shoot grew best at 2mg/L of Cu2+.There is a highest germination rate when the density of copper ions was 4mg/L. When the density of Cu2+ reached 10mg/L, the number of root length and shoot length reduced slowly. And the root could’ t grow nearly.
