摘 要
本試驗(yàn)以白花蛇舌草作為供試材料,采用KT、2,4-D、蔗糖,三物質(zhì)不同濃度和光質(zhì)配比處理,通過四因素三水平正交試驗(yàn)設(shè)計(jì),以篩選出適合白花蛇舌草愈傷組織繼代培養(yǎng)的最佳配方,然后以最佳配方為基礎(chǔ),采用L9(34)試驗(yàn)設(shè)計(jì)He-Ne激光不同輻射劑量、不同輻射時(shí)間及不同光照強(qiáng)度的配比處理,通過對吲哚乙酸氧化酶活性等生理生化指標(biāo)測定,篩選出有利于白花蛇舌草愈傷組織生長的最優(yōu)組合。試驗(yàn)結(jié)果表明:在正常光培養(yǎng)條件下,白花蛇舌草的繼代培養(yǎng)的最佳配方是MS+2,4-D4.0mg/L+KT1.5mg/L+蔗糖30g/L。在He-Ne激光輻射時(shí)間與劑量分別為3mA、3min和光照強(qiáng)度為1500lx時(shí),對愈傷組織的吲哚乙酸氧化酶活性的影響達(dá)到最低;在He-Ne激光輻射時(shí)間與劑量分別為6mA、3min和光照強(qiáng)度為2000lx時(shí),對愈傷組織的可溶性蛋白質(zhì)含量的影響達(dá)到最大;在He-Ne激光輻射時(shí)間與劑量分別為9mA、3min和光照強(qiáng)度為2500lx時(shí),對可溶性糖含量的影響達(dá)到最大。其中He-Ne激光輻射時(shí)間與劑量分別為3mA、3min和光照強(qiáng)度為1500lx時(shí),為最佳組合。
